Abstract:
The Na/H antiporter isoform, NHE-3, mediates the transport of sodium from the lumen
into the proximal tubule cell in exchange for extrusion of hydrogen ions. This transporter
mediates the majority of renal NaCl and NaHCO 3 reabsorption, and thus plays a
pivotal role in the regulation of extracellular fluid volume, blood pressure, and blood
pH. Our laboratory studies the acute and chronic regulation of NHE-3 by hormones such as
angiotensin II, autocrine/paracrine factors such as endothelin, blood pH, and blood
osmolality. Regulation is studied at the level of gene expression, protein synthesis and
stability, trafficking, and phosphorylation, in cultured renal epithelial cells and in
intact animals. The role of tyrosine kinase pathways and MAP kinases as regulators of
NHE-3 is addressed. Knockout mice are used to examine the role of these regulatory
mechanisms in volume and acid-base homeostasis.
