Abstract:

Class I molecules encoded in the major histocompatibility complex present peptides
derived from viral or tumor antigens to CD8+ cytotoxic T lymphocytes (CTL). Class I
molecules also interact with non TcR lectin-like receptors on natural killer (NK) cells
and subsets of T lymphocytes. Our laboratory studies the role that the murine class I
molecule Qa-1 plays in this process. Qa-1 is of interest because it and its human homolog,
HLA-E, interact with CD94/NKG2 receptors on NK cells and as a result inhibit their lytic
activity. We would like to know how this interaction interfaces with signaling through
other inhibitory receptors on these cells and what functional consequences result. The
approaches that we take include examining the class I pathway for antigen presentation,
binding of peptides to class I molecules and the interaction of such molecules with their
receptors, production of transgenic and knockout mice to study in vivo function,
and determining the role of such receptors on T cell mediated immunity.
