Abstract:

The research interests in our laboratory include oxygen metabolism and stem
cell biology. In our studies of oxygen metabolism, we are utilizing the
myoglobin mutant mouse model and the HIF-1 mutant mouse model to evaluate oxygen
metabolism and adaptive mechanisms in the heart and skeletal muscle. Our
efforts have resulted in the isolation of several novel globin proteins that are
induced in striated muscle that lacks myoglobin. We also have a
significant research effort in the field of muscle stem cell biology. We have
localized the transcription factor, MNF, to the muscle stem cell population and
have pursued a gene disruption strategy to evaluate MNF mutant mice. These
mutant mice display a severe growth deficit and have impaired muscle
regeneration. Furthermore, the MNF mutant mice have a severe decrease in the
myogenic stem cell population and these stem cells have impaired cell cycle
progression due to an induction of the cyclin dependent kinase inhibitor, p21.
Our future efforts will include the isolation of the myogenic stem cell
population, a comprehensive analysis of gene expression including the
preparation of a stem cell library and the definition of the potential of these
cells to contribute to alternative lineages. The results of these studies will
enhance our understanding of stem cell biology and result in therapeutic
advances.
