Abstract:
Research in my laboratory concerns cell motility and proliferation during
wound repair. Fibroblasts in resting tissue are quiescent, sessile cells engaged
in minimal biosynthetic activity. After wounding, these cells become
proliferative and migrate into the wound region where they synthesize and
contract a new connective tissue matrix. Once healing has been completed, the
wound fibroblasts become quiescent and regress through apoptosis. Using
three-dimensional collagen matrices, we have developed an in
vitro model that mimics many of the above features of repair including
matrix contraction. We are using the fibroblast-collagen matrix contraction
model to study (1) cell motility and contractility stimulated by
lysophosphatidic acid, platelet-derived growth factor, and transforming growth
factor beta, and (2) mechanoregulation of cell proliferation, especially in
relation to MAP-kinase signaling pathways.
