Abstract:
The Dallas Heart Study (DHS) is a probability-based cross-sectional study over-sampled for African-Americans designed to improve the understanding of preclinical determinants of cardiovascular disease. Through state-of-the art characterization of phenotypes and genotypes, this study is designed to generate and test hypotheses for improved diagnosis and treatment of cardiovascular disease.

1) Gender Differences in Circulating B-type Natriuretic Peptides
Further details are in the applicant’s research proposal along with preliminary data. We tested the hypothesis that BNP and NT-proBNP levels are regulated by sex hormones. Univariable linear regression and multivariable analyses were performed to evaluate the association of BNP and NT-proBNP with sex steroid hormone levels in women, ages 35 to 49. Log transformed SHBG, an indirect measure of the free estrogen to free testosterone ratio, was strongly associated with log BNP and log NT-proBNP.
2) Reynolds Women’s Study
Polycystic ovarian syndrome (PCOS) is a heterogeneous condition with the clinical features or biochemical findings of hyperandrogenism, anovulatory cycles and polycystic ovarian morphology (PCOM). An estimated 20% of reproductive age women have PCOM, and an estimated 10% of the population has PCOS. PCOS is also associated with metabolic factors recognized to increase cardiovascular risk – lipid abnormalities, insulin resistance and obesity. There is conflicting data about the translation of these risk factors to coronary artery disease and the relative contributions of the intermediate phenotypes of hyperandrogenism and PCOM. The Reynolds Women’s Study was designed to examine the associations of subclinical PCOS with subclinical coronary artery disease and coronary artery disease risk factors among reproductive age women. It is a subset of the DHS, women ages 35 to 49, with additional survey information about menstrual cycles, serologic measurements of total testosterone, SHBG, FSH and LH, and MRI data about ovarian structure.
