Abstract:
The primary function of the respiratory system is to provide adequate gas exchange for the organism. Just like other organ systems, genetics has an important impact on many aspects of the respiratory system, from the shape of the thoracic cage to the intrinsic response of the lung parenchyma to various insults. We are using genetic approaches to identify the primary determinants of lung disease, especially diseases of lung parenchyma such as emphysema and pulmonary fibrosis.

Our approach is to collect the medical history of families containing a number of affected individuals. We look for the pattern of inheritance, the phenotypic heterogeneity, and evidence of interactions between specific environmental exposures. Blood samples and additional clinical information are collected from the affecteds and the first-degree at-risk individuals. These provide us with the resources for determining the genetic underpinnings of these diseases.

We have recently found a gene defective in a specific form of emphysema, familial spontaneous pneumothorax, which results in the formation of multiple holes at the periphery of the lung. The specific role of this protein in lung development and its cellular function is a focus of the laboratory.
