DNA manipulation enzymes Enzymes which facilitate the in vitro modification of cloned DNA molecules. They include RESTRICTION ENZYMES for site-specific cleavage of the DNA, DNA LIGASES for covalently joining DNA fragments, and DNA METHYLASES which are used to methylate restriction enzyme recognition sites, thereby avoiding cleavage at these positions by the corresponding restriction enzyme. Although each class of enzyme occurs naturally, their biological roles do not necessarily dictate the uses made of them in DNA manipulation in vitro.
