Okazaki fragment A short fragment of DNA generated on the lagging strand during DNA REPLICATION. Because DNA polymerase can only elongate a polynucleotide strand in a 53 direction, a new DNA strand has to be synthesized discontinuously along one of the strands of the unwound duplex at the replication fork. Each Okazaki fragment is initiated as a short RNA primer and is 1000-2000 nucleotides long in prokaryotes, and 100-200 nucleotides long in eukaryotes. The gaps between fragments are subsequently filled in by DNA polymerase and joined by DNA ligase.
